Ayodele Fayose came into political limelight in 2003 during the tenure of Former President Olusegun Obasanjo, Fayose who contested for the 2003 Ekiti State Gubernatorial Election under the platform of the People's Democratic Party (PDP), defeated the then incumbent Governor, Otunba Niyi Adebayo of the Alliance for Democracy (AD), thereby becoming the First PDP Governor likewise the Second Executive Governor in the State. During the initial period of his leadership in the State, he enjoyed massive goodwill from the poor and downtrodden that he readily identified with. He was a very generous man and grabbed any opportunity to relate with the poor.

In a dramatic twist, crisis hit the State as the peaceful atmosphere became tense, a development that led to the impeachment of Fayose as Governor and the subsequent appointment of Brigadier General Adetunji Olurin as the Sole Administrator of the State following a declaration of State of Emergency by the then President Olusegun Obasanjo. Fayose fled the country after his impeachment, with his removal from office, he had lost constitutional immunity, he was in exile until Obasanjo left power in 2007.
He returned to the country in December 2007 and was well received in Ado-ekiti, the State Capital, a clear indication that he has not lost his support base. In 2011, Fayose contested for Ekiti Central Senatorial Seat under the Labour Party (LP) platform but lost the election to Babafemi Ojudu of the Action Congress of Nigeria (Nigeria). He later returned to his former party, the PDP.
Consequently, he decided to continue his Political career from where he left off and fortunately, he emerged as the PDP Governorship Candidate for the June 2014 Gubernatorial poll in the State, he won the keenly contested election defeating the incumbent Governor Kayode Fayemi of the All Progressives Congress (APC). Fayose was subsequently declared by the Independent National Electoral Commission (INEC) as the new Governor of the State, he was sworn-in in as Governor on the 14th of October, 2014 during the Administration of Former President Goodluck Jonathan.
Since the ruling All Progressives Congress (APC) came into power in 2015, an outspoken politician, Fayose is well-known for his frequent criticism of the incumbent President Muhammadu Buhari and his party, the All Progressives Congress (APC). He successfully completed his four-year tenure in October 2018, making him the only Nigerian Governor who defeated two incumbent Governors and also served under three Presidents, namely Former Presidents, Olusegun Obasanjo and Goodluck Jonathan, likewise under the incumbent President Muhammadu Buhari, a feat that has not been achieved by any other Nigerian Governor till date.
